Arming Pilots Essay After the September 11th attacks on America’s homeland, citizens became aware that action must be taken to protect passengers and pilots from terrorists. Although strict security measures have been put in place at all American airports, the only way to be certain that another terrorist attack does not occur using commercial aircraft lies in arming pilots with handguns. Not only will arming pilots help act as a preventative measure, it will ensure safer air travel and restore faith in America’s main form of long distance travel. Even though the number of hijackings is minuscule compared to the number of flights, arming pilots would be a first line of deterrence and a last line of defense. Some people may argue that other means should be used to stop hijackers, but as is the case with any illegal activity, if a person wants to commit a crime they will find the means. Therefore, the only way to prevent aircraft hijackings is with the use of guns. One alternative way that has been suggested is arming pilots with tasers, electric shock probes. These are considered problematic by many experts and do not guarantee bringing down a hijacker. As Criminal Justice expert Michael Swedemen said, â€Å"Had the pilots on the ill-fated planes on September 11th had stun guns, they might have dropped one of the hijackers, but the remaining three or four still would have overpowered the plane† (Dougherty, 2002, January 22). Arming pilots would also aid airlines in recovering from the economic downfall that has plagued the industry since the attacks. With armed pilots, passengers would know they are safe from any possibility of a hijacking and the only thing that might cause catastrophic damage would be an unforeseen mechanical problem. Should hijackers be among the passengers they could easily be brought down with guns similar to that of a defiant criminal. Opponents have argued that a stray bullet might penetrate the fuselage or inadvertently hit an innocent passenger. The Air Line Pilot’s Association has called for arming pilots using special ammunition that can kill an attacker but not pierce the fuselage (Dougherty, 2002, January 22). Hitting an innocent passenger is a risk that must be taken. One innocent life lost is far better than the possible countless losses on the hijacked aircraft and immanent destruction resulting on the ground. If passengers know that hijackers cannot take control of the plane, millions of Americans would once again begin flying which would improve economic conditions in a hurting industry. Other questions lie in the ability and mental state of a pilot to carry weapons. Senator Bob Smith (R-N. H. said, â€Å"Commercial pilots are trusted with the lives of thousands of Americans every day, and with proper training they could †¦ protect our skies from future acts of terrorism† (Dougherty, 2002, March 19). Pilots are some of the most mentally, physically, and psychologically tested people with hand/eye coordination that is among the best in any profession. Issuing firearms to pilots would, in effect, be like issuing a gun to a police officer. A pilot should be able to defend his aircraft instead of being left defenseless. The second amendment to the Constitution guarantees the right to bear arms. As this is true for a homeowner to protect his family and himself, this should be true for pilots and their aircraft should they come under attack. Although the idea of using air marshals still exists, they are seen as being less effective. Along with ineffectiveness, the cost of placing an air marshal on every flight is unrealistic. A pilot who has undergone the proper training can perform the same task while being paid by the airlines, rather than spending taxpayer’s dollars (Shaw, 2001, September 19). As support for handguns in the cockpit grows, we must ask ourselves if four guns had been in the cockpit on September 11th would 3,000 people be dead, two 110 story buildings have been brought down, the Pentagon attacked, and a nation left in shock? Americans must realize that committed terrorists will be able to get past security, leaving pilots as the last line of defense. Along with pilots, many Americans feel these measures have to be taken in order to make the friendly skies, friendly again.

GENOME is consisting of reads, these reads are very large in amount to manipulate and also to store and maintains. Sequencing machine produce output of short overlapping substrings, these substring are called reads. The sequence assembly reconstructs genome sequence of these reads. These genome sequences are long and continuous. Assembly software for Nest Generation Sequencing (NGS) must be a very accurate, fast and have a less memory consumption. PASQUAL is tool used for faster work of NGS GENOME assembly. For address challenges of NGS assembly, parallel algorithm and compressed data structure are used in PASUQAL. DNA is made from simple unit called nucleotides (nt). Nucleotides having four types namely A, C, G, and T. In sequence start and end are denoted by 5’ and 3’ respectively. Deducing the order of nucleotides from cell and encoding it as a string of letters is called a DNA sequencing process. This process cannot read whole sequence continuously, so it breaks DNA molecules into small part, which is used in chemical reaction as templates to produce short sub-sequences called reads. Major problem is a reconstruct the original genome sequence from reads. For these purpose GENOME assembly algorithms are used. A GENOME assembly uses many automated rounds to improvements, but it inspected and edited by specialists. Assembling reads into a long contiguous sequence is called contigs. The genome sequencing is process of reading sequence of base pairs (bp). Organism genome consists of base pairs, which is derived from two stranded of complementary bases. This is a main part to the study of genomes in bioinformatics. Except Whole – Genome Shotgun (WGS) sequencing machine, no other current sequencing method is capable to read whole sequence in one pass. De novo assembly not uses any reference sequence aids to reconstruction of original sequence, because of these it is used in PASQUAL. We have to generate a large number of reads in a small amount of time, for these purpose we used a Next Generation Sequencing (NGS) technologies. For these purpose scientist uses the capillary electrophoresis (CE) – based Sanger sequencing, scientists able to manifest genetic information for any biological system. Because of these it is adopted by many research laboratories. But it has many limitations like throughout, scalability, speed and resolution to preclude in scientists research study. To overcome from these problem, these is new technology is introduced namely as Nest-Generation Sequencing (NGS), that become a reason for boost in research area in bioinformatics and genomic science. This gives an important breakthrough in fields like human disease and agriculture research. The principle behind NGS is similar to CE. CE generates small fragments of DNA. These fragments are sequentially identified from each fragment, which is re-synthesized from DNA template. NGS perform similar work in parallel fashion, which is population of millions of reaction rather than single or few DSN fragments. Due to this NGS produces hundreds of gigabases of data in single pass/sequencing run. NGS perform its operation as – a single genomic DNA is firstly fragmented into numbers of small segments, which is also known as library of segments. These segments are uniformly and accurately sequenced in millions of parallel reactions. These strings of bases are called as reads. Then these reads are reassembled by tow technique, first is known reference genome called as scaffold (re-sequencing) and second is without any reference genome (de novo sequencing). The output is set of aligned reads represents entire sequence of each chromosome in the gDNA. Individual sequence reads are reassembled by aligning to a reference genome. The Whole–genome sequence is derived from the consensus of aligned reads. NGS output is increased as a rate that outpaces Moor’s law. A single pass can produce up to one gigabase (Gb) of data, at the time of invention i.e. in 2007. At 2011 it reaches up to terabase (Tb) of data in single pass/sequencing run. i.e. almost 1000Ãâ€" increase in four years. Because of this ability of NGS, researchers can move from idea to full data sets in few hours or days. Using CE technology sequencing of human genome takes a time around 10 years. But using NGS we can generate five human genomes at a single run. So it reduces the cost of genome projects. In NGS we can tune resolution of genome experiments. It is possible to produce more or less data, also it support zoom in particular regions of genome with high resolution or view with low resolution but it is more expansive. To do these researchers can tune coverage generated in experiments. This ability gives number of experimental design advantages. Because of various advantages of NGS has permeated in many areas of study. It uses OpenMP for shared memory parallelism, because of its good working between programmer productivity and performance. PASQUAL uses OLC approach and obtain high quality solutions with combination of tailored algorithms. PASQUAL can handle billions of bases. It uses de novo assembly, because of it does not need any reference to produce original sequence. Algorithm constructs biological sequences in parallel by suffix array, and it is good key for parallel performance and memory optimization. Index stage and string graph construction is used for finding overlaps. Misassembles of genome sequence by PASQUAL is significantly less than ny other assemblers. PASQUAL can handle billion of bases in less time, because it uses pipelined stages and compressed data. It has advantages over SOAPdenovo and k-mer like SOAPdenovo is only a tool having comparable speed and k-mer is restricted to smaller length than 128. Rather than PASQUAL produces less errors compared to any other tool. 4. Literature Survey 4.1 De Novo Genome Sequence Assembly In year 2008 to 2012 these are many sequencing techniques are developed, due to these there is major drop in cast from 1/100000th to 1/100000th of price. De novo algorithm is inherited from the SOAPdenovo2 framework. De novo sequencing involves novel genome; it requires specific assembly of reads (sequencing reads). It requires unique combination of length, depth of reads also it requires flexible paired-end insert size. Unpatrolled raw read makes confident and efficient production and long contig assemblies. De novo sequencing assembly is preferred for study of non-model organisms, because it is cheaper and easier to construct a genome. The reference-based assembly uses mapping on to reference genome, because of these it has inability to account for incidents of structural alteration of mRNA transcript. De novo assembly provides means to discover new and unknown sequence in biological research. Reading of whole sequence at once is limited, de novo methods are irreplaceable. It mostly used to discover new and unknown sequences, which is important in biodiversity in world. 4.2 Overlap/Layout/Consensus (OLC) Approach Overlap Layout Consensus (OLC) method is used in de novo assembly. It has a three steps overlap, layout and consensus respectively. In overlap stage graph is constructed, graph is made up of basic assembly. In layout stage this given graph is compressed. And in the consensus stage upon graph data, genome sequence is determined. These data is generated in previous two stapes. Overlap:- In the overlap stage, each and every reads are compared with every other read, and these is perform in both direction forward and reverse complement orientations. It is very time consuming procedure especially in set of large reads. Layout:- Finding path in OLC graph in not an easy task, because it has million of nodes and edges, and it very tedious task to find path that visit each node exactly ones. In this stage it OLC assembly graph is simplified, where assembly graph (i.e. segments) are compressed into contigs. Consensus:- This is a final stage of OLC approach, at this step assembly graph is reduced to large scaffolds i.e. single scaffold. It start from left most read of each scaffold, OLC algorithm computes consensus of all the reads composing each scaffold. Gaps in the genome may still be presents if the consensus step had insufficient mate-pair or repeat contig information. If an assembly had gaps, it would result in a fragmented genome, composed of multiple scaffolds because the gaps between the scaffolds could not be joined. 4.3 Shotgun Sequencing Sanger DNA sequencing technique work on limited distance in sequencing primer from 30 to 350 nt i.e. read length. Because of chain termination very few product can produce chain. These work at best ability to sequence maybe 500 bases a day and it is infeasible for human genome which have billions of bases. Another approach is, first divide DNA in to smaller fragments which is individually sequenced. Then these fragments are reassembled into original form based on overlaps. This strategy is known as shotgun sequencing, it also known as shotgun cloning. In shotgun sequencing, it randomly sheared into small pieces (usually about 1kb) and sub cloned into universal cloning vector. The library of sub fragments is sampled at random, and sequence reads are generated. These reads are assembled into contig. From this procedure complete sequence of clone generated. Shotgun technique can identify gaps (i.e. there is no sequence available) and single standard regions (where there is sequence for only one stand). They are targeted for additional sequencing to produce fill sequenced module. 5. Full Stage Pipelining and accuracy in PASQUAL 5.1 Motivation for this topic With an explosive growth of genome research area and in genome sequencing data, there is huge demand for tool and systems that enables researchers to more efficiently and more effectively work. NGS technology can produce shorter reads as compared to previous sequencing and delivers higher coverage. Coverage means ratio of total length of reds to genome length. Typically NGS generates reads from millions to few billion. This result is depending upon genome size and coverage. Due to high improvements in technologies, data sets to grow larger. Use of the Mock-epic Style in The Rape of the Lock "The triumph of the Baron's rape is in exactly the same high language as it would be if he were Hector." In The Rape of the Lock, Pope uses the mock-epic style to satirise the seriousness with which a trivial misdemeanour (the theft of a few strands of hair) and the ways of gender polarised society can be blown beyond all sense of proportion. Thus the male mentality, through the Baron, is portrayed as lacking depth or personality beyond that required to achieve its ends; men objectify and devise "strategems" (4,120) to conquer their female obsessions; they are "victor[s]" (4,162) who self-importantly congratulate themselves as meriting "wreaths of triumph" (4,161) when they have seized what they desire. The Baron claims that the "glorious prize" is his in perpetuity, whilst many conditions which will never be fulfilled ("while fish in streams, or birds delight in air" 4,163) remain unfulfilled. In this satirising of the epic mould such trivial occurrences are substituted in place of truly fantastic possibilities (mighty cities falling, for instance) for the purpose of putting the lock's severing into a more realistic perspective — this is made even more explicit in the following canto (4,8 "[no-one ever] felt such rage, resentment, and despair / as thou, sad virgin! for thy ravished hair" — meaning that perh aps Belinda over-reacts, in Pope's opinion, just ever-so slightly.) He also then reinforces his satire with a broadening of humour, and a stab in the direction of then-popular culture: specifically, "Atalantis" (4,165) was no great enduring writing but a cheap, scandalous work of fiction, "notorious for its thinly concealed allusions to contemporary scandals", pe... ...rder of life.") Obviously the ultimate aim of the poem is to mitigate the severity of the liberty taken in the theft of the lock (as seen in the minds of those involved in the familial dispute.) Mock epic assists Pope in achieving this without being seen to trivialise the assaulted feelings of the victim — the high language and drama of his work accords to the act of the lock's severing a grossly inflated significance, which retains enough of its epic origins not to be viewed as derisive sarcasm. As a satirist Pope is therefore presenting for the appraisal of his readership the notion that the loss of the lock does not deserve the intensity of ill-feeling which has resulted from it. One flew over the cuckoo's nest One flew over the cuckoo's nest One Flew Over the Cuckoo's Nest For as long as time could tell, whenever and wherever there is a corrupt ruling system in place, there will always be an opposing force trying to over throw it. This ruling system can be a variety of things. In some cases it is the government, a boss, or basically anything or anyone that has some type of control or authority over something else or someone else. In some cases the opposition can successfully take over control of these corrupt systems, while in other incidents the opposition is pitifully pounded back to silence. In other cases, the opposing force will be beaten, but in their shadowy remains lye a path for future generations to follow. In the case of Mc Murphy and the Big nurses a power struggle, the opposition (Mc Murphy) gets beaten silent, yet his words will continue to ring throughout the halls of the ward. Mc Murphy has been made a martyr, and has ultimately stripped Big Nurse of her abused powers and paved the way for fellowmen to escape her entrapment. Based on the novel One Flew Over the Cuckoo's Nest by Ken Kesey, it seems that the authors' perspective on this issue is that the system in place during this time period is in need of change. Ken seems to like the revolutionist characteristics found in his main character, and emphasizes the idea of questioning the authority power. His belief seems to be that even if you are not successful in changing the system in entire, the effects of a person trying can still be very effective. In fact, if all you manage to accomplish is changing a small aspect of the system, it was still all well worth the fight. For as seen, the effects of a minor victory, ripple into much larger victories in the battles to come. As a revolutionist, you set an example for others to follow and in essence pave the way for others to follow. In One Flew Over the Cuckoo's Nest, the main character, Randle Patrick McMurphy, fights to change the system in a mental hospital. McMurphy is very outgoing, loud, rugged, manly, a leader and a rebel. From the first couple scenes of the book, there is a constant power struggle between the patient's new found savior McMurphy, and the evil Nurse Ratchet who rules their wing of the hospital with an iron fist.

Let's call it reality because Brown was so deeply affected by it. So, the first question is answered, now, was the reality created by the devil to convert Brown or were all the people Brown encountered really as evil and hypocritical as they appeared. After Brown left his wife, Faith, he started on a "dreary road, darkened by all the gloomiest trees of the forest. . ." He was feeling not so alone, and thought that the devil himself could be at his back. At this very instant, Brown saw a figure leaning against a tree. The devil came as soon as Brown called him. Why was he so afraid he might meet the devil, when it seems to me he know he was going on an evil journey with the devil? The fellow traveler (let's just call him the devil) shared a resemblance to Brown. In fact, when goody Cloyse sees him she say he is "in the very image of my old gossip, goodman Brown,. . ." So, the devil contrived himself to look like Young Goodman Brown, so Brown would feel a sort of brotherhood, or fatherhood, to the devil. He made himself more like Brown so Brown would more easily be swayed. To me, if the devil can change his appearance, what would stop him from changing others appearances. Who's to say that it was actually goody Cloyse that Brown saw . Brown is so hurt that that the woman who taught him his catechism could actually be a witch. As would be anyone who's idea of what is right and wrong is shattered. But Brown wasn't so easily swayed as the devil had hoped, Brown still resisted. So, the devil thought he would shake Brown even more. He contrived the voices of the deacon Gookin and the minister.
